چکیده

Stokes (1944) reported the replaceability of folic acid by thymine for Streptococcus lactis strain R as well as other folic-acid-requiring streptococci, and Fink et al. (1954) noted that high concentrations of recrystallized dihydrothymine supported growth of Streptococcus faecalis (ATCC 8043) deficient in both folic acid and thymine. The question arose, however, as to whether the latter response might be due to trace amounts of thymine present in the dihydrothymine, and it was difficult to rule out this possibility by ordinary analytical methods. The present paper describes the results of a microbiological test for residual thymine-like activity following subjection of preparations of dihydrothymine to mildly alkaline conditions under which thymine was stable but dihydrothymine was hydrolyzed to jl-ureidoisobutyric acid (Fink et al., 1954; Batt et al., 1954).

Evidence against the folate-mediated formylation of formyl-accepting methionyl transfer ribonucleic acid in Streptococcus faecalis R.

The formylation of formyl-accepting methionyl transfer ribonucleic acid (tRNA fMet) was investigated in Sfreptococcus faecalis R, an organism incapable of synthesizing folic acid but whose growth does not require the vitamin under deiined conditions of culture. These studies were carried out because it was not clear how an organism with no folate could initiate protein biosynthesis. Growth of S...

Regulation of folylpoly-gamma-glutamate synthesis in bacteria: in vivo and in vitro synthesis of pteroylpoly-gamma-glutamates by Lactobacillus casei and Streptococcus faecalis.

Lactobacillus casei and Streptococcus faecalis accumulated labeled folic acid and metabolized this compound to poly-gamma-glutamates of chain lengths of up to 11 and 5, respectively. Octa- and nonaglutamates predominated in L. casei, and tetraglutamates predominated in S. faecalis. The most effective monoglutamate substrates for the L. casei and S. faecalis folylpoly-gamma-glutamate (folylpolyg...

The influence of folic acid and vitamin B12 on nucleic acid metabolism in microorganisms.

A combination of purines and thymine has long been known to substitute for folic acid (FA) in the nutrition of microorganisms requiring the vitamin. The purines were found to be interchangeable, but the effect of thymine was specific and could only be duplicated by its desoxyriboside, thymidine (l-4). From these and other studies on inhibition analysis (5, S), it was concluded that the purines ...

Microbiological assay for thymine using a mutant of Streptococcus faecalis.

Doctor, V. M. (University of Texas, Houston). Microbiological assay for thymine using a mutant of Streptococcus faecalis. J. Bacteriol. 84:17-22. 1962.-A rapid microbiological assay procedure for the determination of thymine was devised. The test organism, Streptococcus faecalis Akt, a mutant of S. faecalis ATCC 8043, gives one-half the maximal growth response to 1.6 mug of thymine (assay range...

Biosynthesis of thymine and formyltetrahydrofolate by vitamin B12-deficient cells of Lactobacillus leichmannii.

A relationship between the functions of vitamin Blz and folic acid (pteroylglutamic acid) is implied by the hematological response induced by either of these vitamins in patients with pernicious anemia (1,2). This interrelationship between the two vitamins may arise because of separate functions at different steps in the formation of common products, such as thymidine, or because vitamin Bit ha...
